HISTOMORPHOLOGICAL STUDY OF THE EFFECTS OF PHOTOPERIODISM ON THE ADRENAL GLAND OF AFRICAN GIANT RATS (Cricetomys gambianus)

Abstract
The study investigated the effects of photoperiodism on the adrenal glands of African giant rats (AGR)¬. Thirty captive AGR of both sexes were divided into 3 different groups of 10 rats each (5 males and 5 females). Group 1 had equal hours of darkness and light for 8 weeks, Group 2 had total darkness for 8 weeks while, Group 3 had total lighting for 8 weeks. Each rat was euthanized using sodium pentobarbital at 86mg/kg BW IP. Grossly, Group 2 rats showed massive deposition of adipose tissue. The relative weight of the adrenal gland at 8 weeks increases significantly (P<0.05) in Group 1 and 3, while, the mean length increases significantly in Group 1 and 2. Histologically, the adrenal gland showed 3 different layers (capsule, cortex and medulla) across the groups. The adrenal capsule was composed of smooth muscle fibres, blood vessels and nerve ramifications, while, the cortex showed 3 distinct zones (zonal glomerulosa, fasciculata and reticularis). The zonal glomerulosa was composed of small columnar cells arranged in tubular fashion, zona fasciculata was composed of parallelly arranged cuboidal cells and zona reticularis was composed of network of anastomosed cells surrounded by fenestrated capillaries. The adrenal medulla comprised of irregularly arranged anastomosed round or polygonal shaped cells with basophilic cytoplasm. The group exposed to total light shows elaborate zona fasciculata, zona reticularis and medulla when compared to those exposed to total darkness. Exposure of AGR to continuous light and darkness significantly increases the relative weight of the adrenal gland with a decrease in adipose tissue deposition and increases the length of adrenal gland with an increase in adipose tissue deposition respectively.
