PRODUCTION AND PURIFICATION OF CHOLESTEROL OXIDASE FROM SOIL STREPTOMYCES OBTAINED FROM MUNGADI FADAMA LAND, KEBBI STATE, NIGERIA

Abstract
Cholesterol oxidase is an alcohol oxidoreductase flavoprotein with wide biotechnological applications. In recent years, microbial cholesterol oxidases have gained great attention due to its widespread application in many industries. Soil sample was collected from Mungadi fadama land area and Streptomyces was isolated using starch casein agar media. The isolates (Streptomyces) was identified using their physical and microbial characteristics and screened for the potentiality of producing cholesterol oxidase using red coloring stain method, then inoculated into production media for the production of cholesterol oxidase. After production period, the protein content of the culture medium was determined as well as the enzyme activity using modified Yazid method. The enzyme was partially purified using ammonium sulphate method. The morphological and microbial characteristics showed by the selected isolates (Gram positive, earthy ordour, white in colour, rod shaped and no motility) shows that, the isolated bacteria were Streptomyces as described in standard Streptomyces project. The activity of the enzyme was 0.92 U/mL with control 0.11 u/ml. The specific activity was 83U/mg with yield 29.64% after one step ammonium sulphate precipitation. Bacterial Streptomyces from Mungadi Fadama soil was capable of producing cholesterol oxidase enzyme and the purified enzyme can be used for many industrial applications.
